SUMMARY AND RECOMMENDATIONS Data integra*on, analysis, and interpreta*on of eight academic CLARITY-BPA studies JJ Heindel, S Belcher, JA Flaws, GS Prins, SM Ho, M Jiude, HB PaCsaul, W Ricke, CS Rosenfeld, AM Soto, FS vom Saal, RT Zoeller

  1. SUMMARY AND RECOMMENDATIONS Data integra*on, analysis, and interpreta*on of eight academic CLARITY-BPA studies JJ Heindel, S Belcher, JA Flaws, GS Prins, SM Ho, M Jiude, HB PaCsaul, W Ricke, CS Rosenfeld, AM Soto, FS vom Saal, RT Zoeller Reproduc*ve Toxicology , July 2020 Jul 12:S0890-6238(20)30150-7. doi: 10.1016/j.reprotox.2020.05.014

  2. CLARITY CLARITY-BP -BPA : Comb mbined Guideline AND AND Academi mic studies to inform m risk assessme ment on BPA safety UR0GENITAL TRACT GUIDELINE Weight Histology ACADEMIC Urethral Colliculus enlargement Urethra morphometrics Body weight THYROID GUIDELINE Weight Histology ACADEMIC Thyroid hormone levels Thyroid hormone brain endpoints Adapted from Pat Hunt

  3. REPRODUCIBILITY • High • Variation • experimental design • model • time points CLARITY findings: conclusions remain the same

  4. SENSITIVITY Affected organs: Mammary Heart Prostate Uterus, Ovary Urethra Brain • Reproducible endpoints • Far more sensitive Ideal for future EDC testing paradigms

  5. LOW-DOSE and NON-MONOTONIC EFFECTS • Significant adverse effects at the lowest dose 2.5 µ g/kg BW • Consistent non-monotonic (i.e. non linear) responses • Integrative mixOmics analysis: strong, complex inter-relationships Strong evidence: low-dose effects are NOT spurious!

  6. STUDY STRENGTHS ü Animals treated at FDA under GLP guidelines ü Investigators fully blinded ü Academic data locked before codes released ☞ All Academic studies meet requirements for inclusion in risk assessment analysis

  7. Prior to CLARITY … Guideline conclusions: only high doses are toxic 50,000 µ g/kg 50 µ g/kg/day LOAEL = lowest observed adverse effect level; ATI = acceptable tolerable intake, aka RfD = Reference Dose NOAEL = no observed adverse effect level (calculaCon); MTD = maximum tolerable dose; Adapted from Laura Vandenberg

  8. CLARITY-BPA … New LOAEL is unequivocally 2.5 µ g/kg … thus reference dose needs revisiting. 2.5 µ g/kg ? LOAEL = lowest observed adverse effect level; ATI = acceptable daily intake, aka RfD = Reference Dose NOAEL = no observed adverse effect level (calculaCon); MTD = maximum tolerable dose; Adapted from Laura Vandenberg
