  5. BACKGROUND In sillico analysis (blast NCBI excluding all clinical species ) One putative candidate identified

  6. Presentation of Zhongshania aliphaticivorans • Z. aliphaticivorans : new species identified in 2013 from low sea tides sediments on the west coast of South Korea. • Gram negative rod • Halophile (grows on Marine Agar) • Facultatively anaerobic, motile • Aliphatic hydrocarbon-consuming bacteria : putative pollutant-degrading bacteria

  7. 2- Presentation of Zhongshania aliphaticivorans • Genome published recently (2016) • No mobile element associated with the M β L gene either B1-M β L Cold-shock Aceyl-CoA hyp hyp hyp bla ZHO-1

  8. ZHO-1 metallo- β -lactamase • 51% amino acid identity with DIM-1 IMP-1 and KHM-1, respectively Kim et al, 2011 • Protein of 246 amino acids

  9. ZHO-1 metallo- β -lactamase

  10. ZHO-1 metallo- β -lactamase

  11. ZHO-1 metallo- β -lactamase • Conservation of amino acid residues involved in the zinc ions binding:  H 120 , H 122 , H 189 , C 208 , H 250

  12. Z. aliphaticivorans susceptibility testing Isolate susceptible to all beta-lactams

  15. Enzyme purification: Purification of the enzyme using ÄKTA-prime (GE Healthcare Life Sciences) • ZHO-1 enzyme: - isolectric point: 6,4 - 246 aa - 26.72 kDa  Purification using Q sepharose column with Tris-HCl pH 8,2 and Tris HCl pH 8,2 ; 1M NaCl

  16. Enzyme purifications: 4L of culture Sonication Nitrocefin assay ÄKTAPrime system SDS-PAGE 12%

  17. Enzymatic activity of ZHO-1 Enzymatic activity using Hepes buffer supplemented with ZnSO 4 (50µM)

  18. Summary: • We identified 1 new class B carbapenemases from environmental isolates from the sea • This carbapenemase confers resistance to many β -lactams including carbapenems • Remaining questions:  Why this metallo-beta-lactamase genes are present in environmental isolates ?  Metallo- β -lactamases in marine environment ? • Other species carries putative B1-M β L : - Cellvibrio sp. - Gilvimarinus agarilyticus
