ESR6: Proteomic analysis for monitoring chronic inflammation Objectives 1.Develop the methodology to access low abundant proteins 2.Develop protocols for standardizing samples for reproducibility over time. 3.Identify modifications of relevant targets which may indicate different stages of inflammatory processes and start quantification of specific targets by MS. 4.In cooperation with ESR 3, this ESR will analyse post-translational modifications and quantities of S100A9 (two translational start sites, phosphorylation at threonine 113). Parallel Project • S100A12, a calcium-binding ‘DAMP‘ is over-expressed in inflammation. • Exists in monomer and multimer forms. • Investigating complex formation and the effect on TLR4 binding using basic cell immunology and proteomic methods.

  3. Next Plans • Complete proteomics goals 1 and 2 – Facilitated by training to be obtained at Dublin University (Research partner in MIAMI project) – To include analysis of samples & continuation in Münster – ? Role with ESR13
